Inclisiran for High Cholesterol, Familial Heterozygous "The drop in my total cholesterol number was dramatic the first 90 days. Dropped from about 280 to 155. My LDL dropped from something like 90 to 55. Won't have the second blood work until two months from now. Side effects: The most aggravating side effect was not diarrhea, but overly soft stools. The overly soft stools cause big-time hygiene problems. This side effect started about two weeks after the shot and continued for about a month. After the second shot, the very soft stools began around two weeks following the shot and continued off and on for a few months. Very, very aggravating. I may have had at times a slight increase in having to urinate. Barely noticeable, but it did happen, and I can't say exactly when that began or tapered off. Haven't had (so far) any undue muscle aches or stiffness."

Inclisiran for High Cholesterol, Familial Heterozygous "I've just had my second injection. Cholesterol dropped, so that's a positive. I still have an injection site rash, possibly due to this last injection being very uncomfortable, unlike the first one. My IBS has been really bad this time too, but not sure if connected. Jury's still out as to whether I would have a 3rd."

Inclisiran for High Cholesterol "Overall good experience with this drug, not had any side effects, I did lose a bit of weight but that's mainly because I went on a low fat low carb diet. The only complaint I have is regarding the area where the needle and the medication go in... it's a bit painful and bruised for a couple of days. Second jab done today hope it does its job and I'll be taken off it in the near future."

Inclisiran for High Cholesterol, Familial Heterozygous "I am a medical professional. I am 78 y/o. Past history of CABG X3 at age 40 in 1987, total cholesterol of 385 and high LDL and low HDL. All numbers are now substantially lowered. Chol now 165. LDL-C is 76, was 165. I haven't noticed any new problems as far as side effects after my first two injections. I have a previous history that includes sciatic distribution pain and discomfort due to mild scoliosis, cervical and lumbar spinal stenosis, and disc problems. Chronic pain controlled presently with Naprosyn and Tylenol ES. Have had no injection site problems or any other obvious new onset problems. I am 78 years old and pretty active. As with every statin I've ever taken, I always wonder whether I am having age-related loss of libido and ED vs. side effects due to medication. I cannot differentiate any new musculoskeletal pain or discomfort with current pain present long before the first Leqvio injection. I have had the first two prelim shots with the next one due in 6 months."

Inclisiran for High Cholesterol, Familial Heterozygous "Started taking first shot March 2 shot June. It is, I believe, responsible for incredible muscle. Stephen is in pain especially in my back and my ribs. It did, however, lower my numbers quite a bit, so I’m grateful for that, but I am not sure it’s worth it if I’m going to be in such severe pain. I hope that the pain goes away in a few weeks when it works through my system."

For High Cholesterol "I had my first injection in the 1st week of April. On the second week of May, my right foot was slightly swollen, and my joints in my toes were very stiff. Unfortunately, I tripped on something quite small while cleaning out my garage. I did not fall, but I fractured my third toe. It is finally healed after 5 weeks, mostly wearing a brace-like shoe. I am now resuming normal walking. However, at the beginning of the second week in June, I lost my balance and simultaneously noticed how stiff my right arm and hand were, so much so that I couldn't effectively break my fall. I fractured my wrist from that experience. I will be in a brace-like mechanism for four weeks. I am due in 4 weeks for another injection. Not at all sure I will continue the treatment."

Inclisiran for High Cholesterol, Familial Heterozygous "This is in regards to side effects of Leqvio. Had the first injection about five days ago. No pain or redness at the injection site. A bit of soreness at the injection site for about 24 hours. A lethargic feeling off and on since the injection. A small amount of muscle or joint pain in the first 72 hours after the injection. No diarrhea as yet. Slightly loose BMs. No fever. Have experienced periods of being 'light-headed', i.e. feeling faint. Have not communicated this to my physician yet. Will do so later this week. I am hopeful this injectable medication will lower my LDL and thus, my total cholesterol number."

For High Cholesterol "Had first injection of inclisiran September 2024. Twelve hours later, suffered stomach cramps, had to sit on the toilet for two hours! Felt bloated and sick, could not eat, loose stools. This lasted five weeks. Digestive system has not been right since. Have craved sugary foods. Now have joint pain in knees, hands, and feet. Severe pain that left me doubled up, now diagnosed with gallstones. It did lower LDL by half, but the side effects are too much. Still not right and coming up to six months after injection. Consultant kept telling me there are no side effects! Glad I am not making it up as has been implied. There has to be quality to life, not just quantity."

For High Cholesterol "I've just had my first injection. I've never been able to tolerate statins, after a triple bypass in March 2025. Inclisiran was presented as a miracle drug. But I've developed all sorts of joint pains, RLS, and insomnia. Having read all the comments, I'm wondering if the injection is responsible."

"I was okay for the first month of the vaccine, and now fatigue, cramp, and UTI. I could not tolerate any of the statins, even in small doses, but while my good cholesterol was good, my LDL was bad! While I have had migraines in the past and cramps in my leg, it seems worse. So I am unsure if the side effect of fatigue is old age, as I have never been old before, lol. I am a 78-year-old woman who is fit and well, apart from this problem with familial cholesterol. The main pain is the price, $1,449!"

For High Cholesterol, Familial Heterozygous "After a >99% occlusion of my LAD was detected, I had a stent implanted at 43 years old. Untreated, my total cholesterol was at 360 mg/dl, with LDL 260. Atorvastatin plus Ezetimibe did not lower the numbers sufficiently. Inclisiran was prescribed by my cardiologist in 2021, and 2 weeks after the first injection, my numbers are amazing. LDL has been below 55 mg/dl ever since. No side effects except for the rather painful injection. Amazing. Will undergo coronary radiography in 2024 to assess whether the good numbers translate into disease regression. (I'm from Germany, where Inclisiran was approved in 2020.)"

Inclisiran for High Cholesterol "I was previously on Repatha for a year and began unbearable side effects, so I was changed to Leqvio. About 3 weeks after my first injection, I began having migraine headaches, intense neck and lower back/muscle pain, fatigue, chest pain, and purpura on my right arm. The symptoms are beginning to lessen now that I am three months in and due for my next injection, which I will not be taking. It did keep my cholesterol level down but not as well as while taking Repatha. My triglycerides raised considerably to a high level."

For High Cholesterol, Familial Heterozygous "I've had 3 injections, last one early December. Straight away, it floored me: headache, blurred vision, feeling weak all the time, muscle and joint pain, can't walk far, so much pain in hips, shivers, like I had influenza all the time. Quality of life-I was an energetic person. I had this injection. On a positive side, reduced cholesterol level in half. I'm due for another injection in June 24, not having it. My life has changed so much. I'll take my chances in exchange for the quality of life to return to what it was!"

Inclisiran for High Cholesterol "Just had my 2nd infusion. First one, no issues besides tenderness in arm of injection. 2nd one, I received 2 days after being on a cruise/flying and all kinds of coughing people. Was OK when I went in, but the next morning felt lightheaded and like I had a horrible cold. Hubs started feeling sickly also, so I am not sure if we got some germs on our travel or if the Leqvio was my culprit. Lasted around a week. Onward, I take warfarin for a mechanical heart valve. On a substantial amount (17 mgs) but normally numbers do not change much. During the period of my first infusion, I needed to start adding more warfarin to my daily dosage. After my second infusion, I realized that the dates of my adding extra started in this time period. I have raised my numbers to 20 mg a day, I was on around 17 mg prior. Correlation? Who knows but will consult with the doctor at Mayo Clinic at the end of the month. Curious if anyone else has had these issues..."

Inclisiran for High Cholesterol "In 2022, I got my first Leqvio injection in the doctor's office. I loved the results, my HDL cholesterol went to normal for a long period of time. This is the only medicine my body tolerated without any secondary effects. I beg my doctors to please get me back on, they prescribe, but insurance declined, subject to a letter of necessity, but that fails to fulfill the purpose. The doctor's office claims they did submit insurance claims, they never got it. I still need this injection. I have conventional Medicare Part B and D."

For High Cholesterol "About to have my third injection. First two did not lower my cholesterol levels very much but told it can take up to a year to respond. Side effects: irritable cough, but this could be from other medicines too (ACE inhibitors). Feeling very lethargic. I have also put on weight. My weight has previously remained stable for years. Has anyone else been troubled by weight gain? Will continue and review after third dose."

For High Cholesterol, Familial Heterozygous "First dose in January. Injection site was painful for a good two weeks. Glad it was in my thigh. Two weeks post shot, I have been having trouble sleeping. I feel as though I've drank ten cups of coffee and am jittery. I actually quit drinking my one cup of coffee in hopes that it would solve the problem, but it did not. I guess I am not sure if it is the shot or something else."

Leqvio has lowered my cholesterol levels to normal with the 2 injections! I will be continuing with these injections as prescribed. Very happy with the results. I am 70 and a female."

Severe body aches gradually faded after the first shot in about 4 to 5 weeks. I am not far enough along from the second shot to know if/when the body aches will fade."

Inclisiran for High Cholesterol "The Leqvio injection was a brief stop at the cardiologist's office. Typical, brief shot in the back of the arm. Easy. They held me there for ~15 minutes to ensure no injection symptoms. The nurse administering the injection did it very well. No pain or discomfort. I don't have serum numbers yet so I am unable to speak to the effectiveness, but I wish all my meds could be this easy."

The shot was relatively painless, less than the flu shot. No side effects. I am a 61-year-old man who feels relieved not to have to take a statin every day."
