"Wow, this stuff is amazing! It did not work at first, but my sister, who is a nurse, told me, that because I had gastric bypass, that I probably need to take more than the 15 mg because gastric bypass patients do not absorb nutrients very well at all and need supplements. I am now taking 45 mg per day without any side effects. And the negative thinking has all but disappeared. I had a DNA test done as my new psychiatric nurse practitioner, after having listened to my woes of having no success with meds or ECT immediately insisted that I take the DNA test. As she suspected, I have not one but two mutations on my MTHFR gene, which decreases my ability to process folic acid in a big way. She suggested that I try Inositol/Choline too."

"Very good supplement for the treatment of depression. It's definitely good as an add-on to prescription antidepressants. Most people with mental health issues are lacking in folic acid and l-methylfolate is the most potent kind of folate."

"So, here goes, I started taking Duplin about five years ago, after I found out I was missing sets of both genes. It really explained a lot of what I went through as a teen. I noticed a life changing difference. I have to explain that I am a young 60 years old. This may or may not hit a nerve with you but I serious think that this drug has put my libido in overdrive. Can anyone relate? I am going to cut them in half from now on and see if this helps."

Deplin (l-methylfolate) "I was so hopeful hearing about the possibilities of Deplin and depression. I've tried twice once at 15mg for about a week and had bad headaches. My doctor suggested taking half (7.5mg) the dose to see if my body would adjust at a lower dose and unfortunately it hasn't. In early start though my mood was better and I was sleeping restfully while the short time on it. The headaches are like migraine severity. Starts in the neck up the back of my head and towards my ears. Lasts for a few days in which I have to Aleve just to be able manage the pain. It would have been great to see if this would continue to improve my depression but it's definitely not worth the painful side effect I experience."

"I have been on this med since March 2017. At first I didn't think much of it. Until recently, I ran out for about a week and was lazy to run to the hospital's pharmacy to retrieve. I can't tell you how much of an effect this will have if you have depression and major depression. I was also checked for the gene mutation where you lack folic acid. I guess this was the cure? It definitely helps and u have to be consistent. I do also suffer from migraines and once in a while Ill get those postdrome symptoms. If this was given to you please make sure to take. There are some good benefits friends."

Deplin (l-methylfolate) "Deplin may have saved my life from menopausal depression. I am heterozygous for the C677T polymorphism in the MTHFR gene, but never had mood issues until late peri menopause (2023). In late March, throughout April and half of May, I experienced serious mood changes related to lower endogenous estrogen. Despite having an estrogen + Dr and good HRT, my mood deteriorated to the point where I went from highly productive and functional to extremely lethargic, unmotivated, exhausted and deeply emotionally vacant. I felt like a shell of myself. I told my new psychiatrist about the genetic test from 2018 and he recommended Deplin 15mg in addition to the Lexapro I had started a year before. I was skeptical, but about 3 weeks after starting it, I noticed a significant improvement in my mood, sleep, energy and motivation and it remains. If you’re in menopause and having difficulty with your mood, you may want to ask your Dr about Deplin and/or the genetic test."

"Soon after beginning menopause I began to wake up after only sleeping for about 4 1/2 hours and I couldn't get back to sleep. This became a vicious cycle where I worried that I couldn't get back to sleep which increased the anxiety which kept me from getting back to sleep. My neurologist recommended Nidra Yoga. She also prescribed 15MG of Deplin. Even though I wasn't low in folate nor was I taking an SSRI, the Deplin was just what I needed to keep me more relaxed. I also read Learn to Relax by Eugene Walker and did progressive desensitization exercises while visualizing myself relaxing and getting back to sleep. The Savvy Insomniac also helped me understand that I could get back to sleep if I didn't stress about it."

"So we are still learning about methylfolate and its effects in depression as it's still pretty new (megavitamin) and we're still learning about the role genetics play in the methylation cycle. I noticed a boost after a week on 15mg then it disappeared. I kept taking even though it wasn't really working anymore. Then after about six weeks I got a huge boost and as I was also on Wellbutrin at the time, it was actually way too much serotonin in my system. I think some people need doses much lower than the prescription strength and it can take a while for the full effect to kick in. It can take a little while to get back to a healthy level but it can definitely help boost whatever else you're on. Look out for hypomania if you're bipolar."

Deplin (l-methylfolate) "I started taking Deplin along with my other meds for depressive disorder and CPTSD. It may have boosted my energy but it caused extreme anxiety. I haven’t felt as panicky while out in public in years as I did while taking Deplin. The anxiety stopped once I stopped taking it."

Deplin (l-methylfolate) "I'm tapering off Prozac and am only taking 2.5mg. I prefer not to go on SNRI 's or SSRI's. I asked my psychiatrist what could I try instead. He suggested Deplin 15 and gave me 12 capsules. I have taken 6 so far and the only thing I noticed that it slows my lower GI tract. Hoping for better feelings in the future."

"My psychiatrist prescribed me 15mg of l-methylfolate after discovering I am homozygous for the c677t variant of the MTHFR gene. Within a few days of taking it, I had nerve pain throughout my body. It felt like I had the flu, but I wasn't ill. Everything, even my skin, just hurt. I've gone my whole life not having any issues with my mthfr issue (the only problem I have is OCD/anxiety) so I'm just assuming I'm better off without it. I am considering switching to a regular multivitamin with a normal dose of methylfolate, but no more mega doses for me."

Deplin (l-methylfolate) "I take lexapro 20mg which works well for my anxiety but not quite so well for my depression and I was always tired . I started taking 15mg Deplin and quickly started to feel more energy. I have more motivation now and just feel better in general. I do have the homogenous C667T MTHFR variant and also had a slight folate deficiency before starting."

"Methylfolate has been a great addition to Paxil for me. After about 7 years on Paxil CR, it wasn't working as well for me. Instead of increasing the dose, methylfolate has helped me feel good again. It's important for me."

"I started Deplin after having genetic testing And learning I had gene mutation. After 3 days of 15mg of Deplin I developed a raised red rash on both checks. From day one I noticed A huge difference in my mood and depression. I have tried probably 30 anti depressant over the years. After researching online I plan to get the 7.5mg dose and cut it in half and start at a smaller dose. As for now I am stopping it until my rash clears up once clears up I will start taking half of a 7.5mg tablet. Hopefully I won't get rash from that lower dose. But definitely recommend especially if you treatment resistant depression."

"I have struggled with depression & anxiety practically all my life. My Mother was also chronically and severely depressed, requiring hospitalizations, ECT treatments, multiple medications with little improvement. I have tried every antidepressant on the market, at least TWICE, with either only a brief period of improvement, or none at all. My PCP suggested we screen for the MTHFR gene mutation and found that I do, indeed, have it. I tried L-Methylfolate 15mg daily for 2 months, and wish I could say it made a difference, but it has not."

Deplin (l-methylfolate) "I had been struggling with depression for many years. Had many bad experiences with the usual anti-depressants, SSRIs and such. After doing the gene psych test and finding out I was homozygous for the MTHFR gene, I asked my doctor if I could try Deplin and he said yes. After just three days it felt like my depression had mostly gone away. After three months I continue to feel better still. This drug has really been a miracle for me. And I have experienced zero side effects from it. I take the 15 mg. I take it in combination with 7.5 mg of mirtazapine. I hope this helps somebody who is on the fence about taking Deplin. It is a little bit pricey but for me it has been worth every penny."
