Mitapivat undergoes extensive hepatic metabolism. The systemic exposure of mitapivat is expected to increase with moderate and severe liver dysfunction. Mitapivat should not be used in patients with moderate to severe liver dysfunction.

Moderate Potential Hazard, Moderate plausibility. Applicable conditions: Renal Dysfunction

The effects of renal dysfunction on mitapivat pharmacokinetics were assessed with population pharmacokinetic analyses. There are limited data available in patients with estimated glomerular filtration rate (eGFR) 30 to less than 60 mL/min/1.73 m2 and no data available in patients with eGFR less than 30 mL/min/1.73 m2; caution is recommended in these patients.
