Zongertinib Disease Interactions

Zongertinib can cause severe left ventricular dysfunction. Left ventricular ejection fraction (LVEF) decrease has occurred with this drug. Treatment with zongertinib has not been studied in patients with history of clinically significant cardiac disease or LVEF less than 50% before starting therapy. This may be considered in patients with heart problems. Patients should be advised to immediately contact their health care provider for new/worsening cardiovascular symptoms.

Zongertinib can cause severe and life-threatening hepatotoxicity (including drug-induced liver injury); this may be considered in patients with liver problems. The effect of moderate to severe liver dysfunction (total bilirubin greater than 1.5 times the upper limit of normal and any AST) on the pharmacokinetics of zongertinib has not been studied.

Zongertinib can cause severe and life-threatening interstitial lung disease (ILD)/pneumonitis. This may be considered in patients with lung or breathing problems other than lung cancer. Patients should be monitored for new/worsening symptoms indicative of ILD/pneumonitis and should be advised to immediately contact their health care provider for new/worsening respiratory symptoms.

The effect of moderate to severe renal dysfunction (estimated GFR 15 to less than 60 mL/min) or end-stage renal disease (estimated GFR less than 15 mL/min) on the pharmacokinetics of zongertinib has not been studied.
