Doxylamine / Pyridoxine Dosage

Usual Adult Dose for Nausea/Vomiting

Doxylamine 10 mg-pyridoxine 10 mg delayed-release tablets:

• Initial dose: 2 tablets orally once a day at bedtime
• Maintenance dose: 2 tablets orally once a day to 4 tablets orally 3 times a day
• Maximum dose: 4 tablets/day

Doxylamine 20 mg-pyridoxine 20 mg extended-release tablets:

• Initial dose: 1 tablet orally once a day at bedtime
• Maximum dose: 2 tablets/day

Comments:

• Treatment should be taken as prescribed, and should not be taken on an as-needed basis.
• Patients should be routinely reassessed for the continued need for treatment.

Use: Treatment of nausea and vomiting of pregnancy in patients who do not respond to conservative management

Delayed-release tablets:

• The dose may be continued if symptoms are adequately controlled.
• If symptoms persist into the afternoon of Day 2: 2 tablets orally at bedtime on Day 2, THEN 3 tablets orally 2 times a day (1 tablet in the morning and 2 tablets at bedtime) on Day 3.
• If 3 tablets adequately control symptoms, patients should continue to take this dose.
• If symptoms persist into Day 4: 4 tablets orally 3 times a day (1 tablet in the morning, 1 tablet mid-afternoon, 2 tablets at bedtime)

Extended-release tablets:

• The initial dose may be continued if symptoms are adequately controlled.
• If symptoms are not controlled the next day, the dose may be increased to 1 tablet orally 2 times a day (in the morning and at bedtime).

Precautions

CONTRAINDICATIONS:

• Hypersensitivity to either active component, other ethanolamine derivative antihistamines, or to any of the ingredients
• MAO inhibitors

Safety and efficacy have not been established in patients younger than 18 years.

Administration advice:

• This drug should be taken on an empty stomach with a glass of water.
• Tablets should not be chewed, crushed, or split.

Storage requirements:

• Bottles should be kept tightly closed and protected from moisture when not in use.
• Desiccant canisters should not be removed from the bottle.

General:

• Limitation of use: This drug has not been studied in patients with hyperemesis gravidarum.

Patient advice:

• Inform patients that this drug may cause somnolence, and they should avoid driving or operating machinery until patients are cleared to do so.
• Patients should be instructed to avoid using other central nervous system depressants concomitantly during treatment.
