Tazemetostat may reduce the blood levels and effects of donepezil. Using tazemetostat with donepezil may also increase the risk of an irregular heart rhythm that may be serious and potentially life-threatening, although it is a relatively rare side effect. You should seek immediate medical attention if you experience sudden dizziness, lightheadedness, fainting, shortness of breath, or heart palpitations during treatment with these medications, whether together or alone. Do not consume grapefruit or grapefruit juice during treatment with tazemetostat. Grapefruit juice can significantly increase the blood levels and effects of tazemetostat, which may increase the risk of serious side effects such as bleeding complications, pain, bone pain, tiredness, shortness of breath, excess fluid around the lungs, and development of new (second) cancers.
