Olaparib may reduce the blood levels and effects of levonorgestrel, which may make it less reliable as a form of birth control. Alternative or additional methods of birth control should be used during treatment with olaparib and for six months after receiving the last dose to avoid unintended pregnancy. This is particularly important because olaparib may cause harm to the unborn child or cause loss of pregnancy (miscarriage). Talk to your gynecologist or other healthcare professional for help in selecting an effective method of birth control that is best for you. If you take hormone replacement for menopause, you should contact your doctor if you experience increased frequency or worsening of your symptoms such as hot flashes, vaginal dryness, or abnormal bleeding. It is important to tell your doctor about all other medications you use, including vitamins and herbs. Do not stop using any medications without first talking to your doctor.

Do not consume grapefruit, grapefruit juice, or Seville oranges during treatment with olaparib. Doing so can significantly increase the blood levels of olaparib and increase the risk and/or severity of side effects such as nausea, vomiting, diarrhea, indigestion, loss of appetite, abdominal pain or discomfort, lung problems, and impaired bone marrow function resulting in low numbers of different types of blood cells. You may also be more likely to develop anemia, bleeding problems, or infections due to low blood cell counts. Talk to your doctor if you have any questions or concerns. It is important to tell your doctor about all other medications you use, including vitamins and herbs. Do not stop using any medications without first talking to your doctor.

Grapefruit juice may increase the blood levels of certain medications such as levonorgestrel. You may want to limit your consumption of grapefruit and grapefruit juice during treatment with levonorgestrel. However, if you have been regularly consuming grapefruit or grapefruit juice with the medication, then it is advisable for you to talk with your doctor before changing the amounts of these products in your diet, as this may alter the effects of your medication. Contact your doctor if your condition changes or you experience increased side effects. Orange juice is not expected to interact.
