Usual Adult Dose for Protein C Deficiency
Acute episode or short-term prophylaxis:
Initial dose: 100 to 120 international units/kg intravenously
Subsequent doses: 60 to 80 international units/kg every 6 hours
Maintenance dose: 45 to 60 international units/kg every 6 or 12 hours
Long-term prophylaxis: 45 to 60 international units/kg every 12 hours
Comments:
- Adjust dose to maintain target protein C activity of 100%
- After resolution of an acute episode, maintain trough protein C level above 25% for the duration of treatment.
- During prophylactic use, higher peak protein C levels may be warranted if thrombosis risk is increased (e.g. infection, trauma, surgical intervention); maintaining trough protein C levels over 25% is recommended.
- Adjust the dose according to the individual patient pharmacokinetic profile.
Use: Prevention and treatment of venous thrombosis and purpura fulminans in patients with severe congenital protein C deficiency.
