Hi there -
Simply sleep (active ingred. diphenhydramine) does NOT interact with letrozole, so it's ok to take with it.
Unisom (doxylamine) is in the same drug category as Simply Sleep, so it's ok to take as well.
You are correct, but with any medicatrion, even with letrozole that lowers estrogen levels (OTC, vitamin, herbal) a patient must inform his/her DR., in case of potential side effects and allergic reactions.
What are some other side effects of Simply Sleep?
All drugs may cause side effects. However, many people have no side effects or only have minor side effects. Call your doctor or get medical help if any of these side effects or any other side effects bother you or do not go away:
Feeling sleepy.
Dizziness.
Thickening of mucus in nose or throat.
Feeling nervous and excitable.
Upset stomach or throwing up.
These are not all of the side effects that may occur. If you have questions about side effects, call your doctor. Call your doctor for medical advice about side effects.

Not trying to scare anyone, just being realistic, before I start any supplements, or an OTC medication I inform my DR. just in case.
Have you ever put yourself in a hypothetical situation in order to learn from it and take safety measures just in case.
I have been on medications for 30 years, and being cautious has helped me quite a lot, be open to other opinions, BTW, no one has the absolute truth, I am not competing with you, I gain nothing, nor want to prove anything, just help the person who posted the question.
Have a nice day, masso

From drugs.com:
Other drugs may affect letrozole, including prescription and over-the-counter medicines, vitamins, and herbal products. Tell your doctor about all your current medicines and any medicine you start or stop using.
