Drug interactions
Moderate drug interactions
Moderate:
amantadine triprolidine
Applies to: amantadine and Vanaclear PD (triprolidine)
Using amantadine together with triprolidine may increase side effects such as dry mouth, constipation, difficulty urinating, heat intolerance, confusion, and blurred vision. More severe side effects may rarely include hallucinations, seizures, irregular heart rhythm, and heat stroke. Side effects may be more likely to occur in the elderly or those with a debilitating condition. Talk to your doctor if you have any questions or concerns. Your doctor may be able to prescribe alternatives that do not interact, or you may need a dose adjustment or more frequent monitoring to safely use both medications. You should avoid or limit the use of alcohol while being treated with these medications. Also avoid activities requiring mental alertness such as driving or operating hazardous machinery until you know how the medications affect you. It is important to tell your doctor about all other medications you use, including vitamins and herbs. Do not stop using any medications without first talking to your doctor.
Food/lifestyle interactions

Moderate:
triprolidine Food/Lifestyle
Applies to: Vanaclear PD (triprolidine)
Alcohol can increase the nervous system side effects of triprolidine such as dizziness, drowsiness, and difficulty concentrating. Some people may also experience impairment in thinking and judgment. You should avoid or limit the use of alcohol while being treated with triprolidine. Do not use more than the recommended dose of triprolidine, and avoid activities requiring mental alertness such as driving or operating hazardous machinery until you know how the medication affects you. Talk to your doctor or pharmacist if you have any questions or concerns.
Disease interactions
22 potential interactions Applies to: 18 conditions, including Depression, Heart Failure, Asthma, COPD, and Seizures.
Alcoholism
Major:
amantadine
Amantadine has a major potential interaction with Alcoholism.
Amantadine may precipitate or exacerbate psychotic symptoms. Rarely, suicide attempts and suicidal ideation have also been reported, some of which occurred in patients with no prior history of psychiatric illness who were taking short courses of amantadine for influenza. Therapy with amantadine should be administered cautiously in patients with a history of psychiatric disorders, emotional instability, or substance abuse, and particularly if they are elderly. It may be prudent to refrain from dispensing large quantities of amantadine to these patients to prevent intentional overdose, which is potentially fatal.
Asthma
Moderate:
Vanaclear PD
The triprolidine in Vanaclear PD has a moderate potential interaction with Asthma.
It has been suggested that the anticholinergic effect of antihistamines may reduce the volume and cause thickening of bronchial secretions, resulting in obstruction of respiratory tract. Some manufacturers and clinicians recommend that therapy with antihistamines be administered cautiously in patients with asthma or chronic obstructive pulmonary disease.
Cardiovascular Conditions and Disorders
Moderate:
Vanaclear PD
The triprolidine in Vanaclear PD has a moderate potential interaction with Cardiovascular Conditions and Disorders.
Antihistamines may infrequently cause cardiovascular adverse effects related to their anticholinergic and local anesthetic (quinidine-like) activities. Tachycardia, palpitation, ECG changes, arrhythmias, hypotension, and hypertension have been reported. Although these effects are uncommon and usually limited to overdosage situations, the manufacturers and some clinicians recommend that therapy with antihistamines be administered cautiously in patients with cardiovascular disease, hypertension, and/or hyperthyroidism.

Gastrointestinal Obstruction
Moderate:
Vanaclear PD
The triprolidine in Vanaclear PD has a moderate potential interaction with Gastrointestinal Obstruction.
Antihistamines often have anticholinergic activity, to which elderly patients are particularly sensitive. Therapy with antihistamines should be administered cautiously, if at all, in patients with preexisting conditions that are likely to be exacerbated by anticholinergic activity, such as urinary retention or obstruction; angle-closure glaucoma, untreated intraocular hypertension, or uncontrolled primary open-angle glaucoma; and gastrointestinal obstructive disorders. Conventional, first-generation antihistamines such as the ethanolamines (bromodiphenhydramine, carbinoxamine, clemastine, dimenhydrinate, diphenhydramine, doxylamine, phenyltoloxamine) tend to exhibit substantial anticholinergic effects. In contrast, the newer, relatively nonsedating antihistamines (e.g., cetirizine, fexofenadine, loratadine) reportedly have low to minimal anticholinergic activity at normally recommended dosages and may be appropriate alternatives.
Glaucoma/Intraocular Hypertension
Moderate:
Vanaclear PD
The triprolidine in Vanaclear PD has a moderate potential interaction with Glaucoma/Intraocular Hypertension.

Hyperthyroidism
Moderate:
Vanaclear PD
The triprolidine in Vanaclear PD has a moderate potential interaction with Hyperthyroidism.
Antihistamines may infrequently cause cardiovascular adverse effects related to their anticholinergic and local anesthetic (quinidine-like) activities. Hypotension
Moderate:
amantadine
Amantadine has a moderate potential interaction with Hypotension.
Dopamine agonists may impair the systemic regulation of blood pressure, with resultant orthostatic hypotension at any time, but especially during dose escalation. Additionally, patients with Parkinson's disease may have an impaired capacity to respond to an orthostatic challenge. For these reasons, patients with Parkinson's disease (or restless legs syndrome) who are being treated with dopaminergic agonists typically require careful monitoring for signs/symptoms of orthostatic hypotension, especially during dose escalation, and should be advised of this risk.
Moderate:
amantadine
Amantadine has a moderate potential interaction with Hypotension.
Amantadine may occasionally cause orthostatic hypotension. Patients with hypotension should be monitored for exacerbation of their condition during therapy with amantadine. Dosage adjustments may be necessary, and patients should be advised not to rise abruptly from a sitting or recumbent position.

Liver Disease
Moderate:
Vanaclear PD
The triprolidine in Vanaclear PD has a moderate potential interaction with Liver Disease.
Limited pharmacokinetic data are available for the older, first-generation antihistamines. Many appear to be primarily metabolized by the liver, and both parent drugs and metabolites are excreted in the urine. Patients with renal and/or liver disease may be at greater risk for adverse effects from antihistamines due to drug and metabolite accumulation. Therapy with antihistamines should be administered cautiously in such patients. Lower initial dosages may be appropriate.
Psychiatric Disorders
Major:
amantadine
Amantadine has a major potential interaction with Psychiatric Disorders.
Amantadine may precipitate or exacerbate psychotic symptoms. Moderate:
amantadine
Amantadine has a moderate potential interaction with Psychosis.
Ordinarily, patients with major psychotic disorder should not be treated with dopaminergic antiparkinsonian agents, because of the risk of exacerbating psychosis. Hallucinations and psychotic-like behavior have been reported with dopaminergic medications. In addition, certain medications used to treat psychosis may exacerbate the symptoms of Parkinson's disease and may decrease the effectiveness of these drugs. The use of bromocriptine in patients with severe psychotic disorders is not recommended.
Renal Dysfunction
Major:
amantadine
Amantadine has a major potential interaction with Renal Dysfunction.
Amantadine is primarily eliminated by the kidney. Patients with renal impairment may be at greater risk for adverse effects from amantadine due to decreased drug clearance. Therapy with amantadine should be administered cautiously in patients with renal impairment. Dosage adjustments are recommended for patients with moderate to severe renal dysfunction.
