Usual Adult Dose for Niemann-Pick Disease Type C

Weight 8 to 15 kg: 47 mg orally 3 times a day
Weight greater than 15 to 30 kg: 62 mg orally 3 times a day
Weight greater than 30 to 55 kg: 93 mg orally 3 times a day
Weight greater than 55 kg: 124 mg orally 3 times a day

Dosage is based on actual body weight.

Use: In combination with miglustat, for the treatment of neurological symptoms of Niemann-Pick disease type C (NPC)

Comments:

The pharmacokinetics of this drug have not been studied in patients with estimated GFR less than 15 mL/min.

Administration advice:

For use in combination with miglustat
Administer with or without food.
Swallow capsules whole; for patients who have difficulty swallowing capsules, administer in 1 of 2 ways:
Oral administration: Open the capsule carefully and sprinkle the entire contents into 15 mL of water, apple juice, or soft food (e.g., applesauce, pudding, yogurt). Stir the mixture for 15 seconds. Consume the entire mixture immediately.
Feeding tube administration (nasogastric or gastric tube): Open the capsule carefully and sprinkle the entire contents into 20 mL of water; do not add the capsule contents to other liquids besides water. Stir the mixture for 15 seconds. Administer the entire mixture immediately via feeding tube; flush the feeding tube with 5 mL of water after administration.
Do not save the mixture for later use.
If a dose is missed, skip the missed dose and resume taking the prescribed dose at the next scheduled time.

Storage requirements:

Store at 20C to 25C (68F to 77F); excursions permitted to 15C to 30C (59F to 86F).
Store in original container; protect from light.

Patient advice:

Contact health care provider immediately if urticaria (hives), shortness of breath, persistent cough, or facial swelling develop in response to treatment.
Patients of childbearing potential: Notify health care provider of a known/suspected pregnancy.
