Major Potential Hazard, Moderate plausibility.

Benznidazole is contraindicated in patients with Cockayne syndrome. Cases of severe irreversible hepatotoxicity/acute liver failure with fatal outcomes have been reported after start of metronidazole (another nitroimidazole drug structurally related to benznidazole) in patients with Cockayne syndrome.

Moderate Potential Hazard, Moderate plausibility. Applicable conditions: Bone Marrow Depression/Low Blood Counts

Hematological manifestations of bone marrow depression (e.g., neutropenia, thrombocytopenia, anemia, and leukopenia) have been reported with the use of benznidazole; these resolved after treatment was discontinued. Only under strict medical supervision should benznidazole be taken by patients with hematological manifestations of bone marrow depression. Complete blood count should be monitored; total and differential leukocyte counts are recommended before, during, and after therapy.

The use of benznidazole has not been evaluated in patients with liver dysfunction; caution is recommended.
