1 drug interaction and 6 additional warnings found for the following 2 drugs:
- chlorotrianisene
- pexidartinib
Drug interactions
Moderate drug interactions
Moderate:
chlorotrianisene pexidartinib
Applies to: chlorotrianisene and pexidartinib
Pexidartinib may reduce the blood levels and effects of chlorotrianisene. If you are using low-dose oral contraceptives, you may have an increased risk of breakthrough bleeding and unintended pregnancy. You should discuss the use of alternative or additional methods of birth control with your healthcare provider. If you take hormone replacement for menopause, notify your doctor if your medication is no longer controlling your symptoms or you experience abnormal bleeding. You may need a dose adjustment or more frequent monitoring by your doctor to safely use both medications. It is important to tell your doctor about all other medications you use, including vitamins and herbs. Do not stop using any medications without first talking to your doctor.
Food/lifestyle interactions
Major food/lifestyle interactions
Major:
pexidartinib Food/Lifestyle
Applies to: pexidartinib
You may experience reduced absorption of pexidartinib in the presence of food. Pexidartinib should be taken on an empty stomach at least 1 hour before or 2 hours after food. This will make it easier for your body to absorb the medication. Also, while taking pexidartinib, you should avoid grapefruits and grapefruit juice. Grapefruit and grapefruit juice can raise the levels of pexidartinib in your body and lead to potentially dangerous adverse effects. Do not increase or decrease the amount of grapefruit products in your diet without first talking to your doctor. It is important to tell your doctor about all other medications you use, including vitamins and herbs. Do not stop using any medications without first talking to your doctor.
Minor food/lifestyle interactions

Applies to: chlorotrianisene
Information for this minor interaction is available on the professional version.
Disease interactions
4 potential interactions Applies to: 4 conditions, including Liver Disease, Renal Dysfunction, Bile Acid Synthesis Disorders, and Biliary Obstruction.
Bile Acid Synthesis Disorders
Major:
pexidartinib
Pexidartinib has a major potential interaction with Bile Acid Synthesis Disorders.
Pexidartinib can cause serious and potentially fatal liver injury. No dose adjustment is needed for patients with mild hepatic impairment. A dose reduction (125 mg twice a day) is recommended for patients with moderate hepatic impairment. Pexidartinib has not been studied in patients with severe hepatic impairment. Avoid this drug in patients with preexisting increased serum transaminases; total bilirubin or direct bilirubin (greater than ULN); or active liver or biliary tract disease, including increased alkaline phosphatase. It is recommended to monitor liver tests before initiation of treatment and at specified intervals during treatment. Withhold and reduce the dose or permanently discontinue pexidartinib based on the severity of hepatotoxicity.
Biliary Obstruction
Major:
pexidartinib

Renal Dysfunction
Moderate:
pexidartinib
Pexidartinib has a moderate potential interaction with Renal Dysfunction.
Dose modification of pexidartinib is needed for patients with renal impairment. It is recommended to reduce the dose when administering pexidartinib to patients with mild to severe renal impairment (CrCl 15 to 89 mL/min, estimated by Cockcroft-Gault using actual body weight). The recommended dosage is 125 mg in the morning and 250 mg in the evening with a low-fat meal.
