Using avanafil together with lonafarnib is not recommended. Combining these medications may significantly increase the blood levels of avanafil. This may increase the risk of serious side effects such as low blood pressure; fainting; visual disturbances including loss of vision; ringing in the ears; hearing loss; and priapism (prolonged and painful erection unrelated to sexual activity). Your doctor may be able to prescribe alternatives that do not interact. It is important to tell your doctor about all other medications you use, including vitamins and herbs. Do not stop using any medications without first talking to your doctor.

Lonafarnib should be taken with the morning and evening meals and an adequate amount of water. Do not consume grapefruit, grapefruit juice, or Seville oranges (also known as bitter or sour oranges).During treatment with lonafarnib. Juice from grapefruits and Seville oranges can increase the blood levels of lonafarnib. This may increase the risk and/or severity of side effects such as nausea, vomiting, diarrhea, loss of appetite, electrolyte (e.G., calcium, magnesium, potassium, phosphate, sodium) abnormalities, liver problems, low red and white blood cell counts, infection, and high blood pressure. Talk to your doctor if you have any questions or concerns. It is important to tell your doctor about all other medications you use, including vitamins and herbs. Do not stop using any medications without first talking to your doctor.

Avanafil can lower blood pressure, and combining it with alcohol may further increase this effect. You may be more likely to experience symptoms such as dizziness, lightheadedness, fainting, flushing, headache, and heart palpitations. You should avoid or limit the use of alcohol while being treated with avanafil, and use caution when getting up from a sitting or lying position. You may also want to avoid drinking large amounts of grapefruit juice, since it may increase the blood levels and effects of avanafil. It is important to tell your doctor about all other medications you use, including vitamins and herbs. Do not stop using any medications without first talking to your doctor.
