"I thought when this came out this was going to be my hope . Unfortunately it didn’t work after two weeks I had to take citrate of Magnesia to empty out . This medication is horrible an causes awful painful gas and epressed mood . My Gastroenterologists refused to give me Zelnorm because I have Controlled Palpitations no heart disease she said it would work. So my last option is colon removal because I can’t live like this anymore using ducolax and bowel preps every three days ."

Motegrity (prucalopride) "In 2020 I was hospitalized for an intestinal blockage. At that time they did not want to do surgery because it was a functional issue so the protocol was NPO for several days. After I left the hospital they put me on the Motegrity 2 mg once a day I’ve taken it for two years and I’ve gained over 25 pounds. It does work in the beginning for motility but then after a year it stopped really working I don’t know why bother staying on it for as long as I did it because the only thing that wound up happening is getting a tremendous amount of weight and becoming depressed from it"

Motegrity (prucalopride) "Motegrity does work, but the effect on me is harsh and I have many side effects. I took 2mg about six hours ago. It hit within 15 minutes. Since then I have gone to the bathroom about 15 times. I didn't make it to the toilet a few times. I'm experiencing racing heart, pounding headache, a jittery/shaky feeling, and intense nausea. I'm dizzy and weak and achy. I take many other meds for other conditions and for all I know they've left my body. There's no way I could deal with this every day. Not sure if I'll try it again, definitely not anytime soon."

Motegrity (prucalopride) "I'm on day 3 and so far it works great. Linzess and Trulance worked a week and stopped, so the jury is out for the long term. The only side effects I have experienced are mild dizziness and back pain. And one strange one, which may be the cause of the loss of appetite. Things that should not taste salty do. That started today. Grapes, French toast so far. I hope that goes away with time."

Motegrity (prucalopride) "After suddenly developing abdominal pain and infrequent BMs 4 years ago without consistent relief from diet changes, or OTC products, Or Amitiza and Linzess, Im trying this (and one other which is less effective). This “worked” initially with soft and loose stool, lots of gas, cramping. Added my OTC again but ineffective. Tried the other sample drug, even less effective Back on Motegrity, same experience and have concluded its exacerbating neck, & back pain to intolerable level, possible wrist and ankle pain, fatigue and the blues. Totally interfering with sleep. The only temporary relief I get is swimming or exercising."

Motegrity (prucalopride) "I was originally given Motegrity without trying the others because I had been following all the lifestyle advice typically given to start off. I had printouts of my daily meals, water consumption, fiber intake, macros. I was also already on a modified diet to to allergens. (I like data.) And it was clear movement for water like linzess wasn’t gonna do it. The first two months, I had to take it with Miralax, 1000 mg of magnesium, and senna at 8.6mg? It took all four to go. I went every day but it was explosive and miserable. So she reduced it down to 1mg and now things are manageable (every two days ish), but I’m constantly boiling. Y’all I’m not at menopause age yet. I shouldn’t be sweating like this. But I’ve been in undershirts in 50 degree weather and I sweat enough to completely stink in a few hours. I also regained all the sudden weight I lost prior to Motegrity in about two months (50 lbs, about 1/4 of my body). Im also queasy all the time."

Motegrity (prucalopride) "My daughter has autism and has had constipation all her life. She has always been on medication for her autism. Motegrity has her making a complete bowel movements but her behavior has become erratic. She is anxious and is having compulsive behaviors. I'm going to stop motegrity for a few days and see how she acts. If her behavior improves I know it's the motegrity. A shame because it is helping her. I'm only giving her 1mg."

Motegrity (prucalopride) "Motegrity worked as soon as I started it. It is the only medication that has ever worked, it changed my life. I can eat regularly. I've stopped taking harsh laxatives. I'm regular and comfortable. Best medicine ever!"

Motegrity (prucalopride) "I have been on Motegrity for over a year and it worked great for awhile and lately I have had problems with feeling jittery anxious and depressed. Feeling jittery and anxious is the worse. I have gastroparesis and also on Famotidine Promethazine Lansoprazole and Motillium so it may be a combination or the Motegrity"

Motegrity (prucalopride) "Worked for 3 days and then back to constipation. Also seemed to not mix well with my anti-seizure drugs. My previous seizure drug had caused a particular type of seizure and depression. I haven’t had those problems since I switched off that drug but experienced both in the past week so I am going to discontinue use."

"I've been taking this prucalopride for about 3 months for CIC. The first 2 months were without incident. I was having small bowel movements consistently each day and feeling fine. Then I was diagnosed with GERD, gastritis, and esophagitis. My stomach was hurting constantly and I was always nauseated. Now, I'm down 14 pounds from the constant inability to eat and the nausea. I also have devastating fatigue and headaches. I thought this was all due to other factors but after reading reviews, I'm thinking the Motegrity is causing the nausea, headache, and fatigue problem. Gonna stop taking it to see if I can get some relief and just work on the GERD and gastritis without constantly feeling like a need to puke."

Motegrity (prucalopride) "I was on Linzess for several months. It completely took away the abdominal pain and for several weeks I had severe diarrhea. Then, it settled down and things seemed to improve but a month later, I was having maybe 1 bowel movement a month. I switched to motegrity, it's been 2 weeks of misery. Headaches everyday, abdominal pain, and bowel movements are more frequent but painful. I'm so disappointed. I will stick it out for awhile longer and hope it improves."

"After lengthy ordeal when my insurance wouldn't cover, I finally obtained Motegrity via patient program. I had been using Senna and some Colace for my neurogenic bowel condition. Senna had/has been causing significant gastro discomfort and incomplete empty has been major problem. For my circumstance, Motegrity was an epic fail. Immediate lethargy, mental haze, constant urination, and even worse than these, it actually seemed to stall bowel activity even more. Candidly, my body interpreted it as poison. Not sure where I go from here but it was wasted time and effort and quite discouraging."

Motegrity (prucalopride) "I wanted to share my story as I just took my first dose of this medication tonight. I'm crossing my fingers because nothing else has been effective, and I am currently down to 107 pounds at a height of 5'5". During the lockdown, I was exposed to toxins in a home I had recently moved into. After four months of being there, I experienced a wide range of symptoms, including severe Gastric Emptying Delay and dysfunction in my colon. Whenever I eat, I suffer from severe consequences. At the moment, I'm on my third round of Xifaxan, which costs $2500 for each two-week treatment, and my gastroenterologist suspects it might be Small Intestinal Bacterial Overgrowth (SIBO) as well. Xifaxan helps with reducing inflammation and bloating, but unfortunately, my colon isn't improving. Currently, the only thing I am able to eat without major issues is eggs, and even then, I have to use a daily enema. I also have Eosinophilic Esophagitis (EOE) and have undergone several surgeries involving scopes, biopsies of the esophageal area, and laparoscopic exploratory surgery. As I enter my third year with these conditions, they are getting worse."

Motegrity (prucalopride) "I have Gastroparesis and have been taking Motegrity for two months now. I'm not really sure how it's supposed to work, I thought it would help relieve constipation and make it easier to go to the bathroom. I am suffering for up to 4 days not being able to have a bowel movement. My doctor thinks that I might have a lot of toxicity in my stomach and I have to take a breathing test. I also suffer from headaches and did not realize until I read all the posts that headaches are common with this."

"I suffer from a range of gastro issues, alongside a damaged ileum intestinal valve due to an autoimmune disease. I have one of the most 'sluggish' bowels my GI doctor has ever seen. Managing my condition has always been difficult, diet can only do so much. Laxatives make me sick and ruin my gut. Then came Prucalopride. It works without fail every time, and boy does it work. It doesn't make me feel sick like other laxatives, and doesn't cause that feeling of 'chemical gut'. It does of course cause some fluid swelling while it works. But once the 'deed is done', I feel like a new person. I don't recommend taking it at night, however, as personally, that causes some really uncomfortable stomach aches. You want to take it on an empty stomach or before food to have the best effect, and to get it 'over and done with' so you can get on with your day (be near a toilet!)"

"I have tried a variety of medications. Lubiprostone (Amitiza) works the best, but it still wasn't enough. I also still use Miralax almost daily and a stool softener once or twice a week. Adding prucalopride to these has helped to give me regular, predictable bowel movements. I take 1/2 pill (1 mg) once a day or every other day. So I'm pleased with how it helps with constipation. But I am minimizing the dose because it affects me like an SSRI antidepressant, which I've been on before and really dislike. Like SSRI's, prucalopride increases seratonin, which is helpful in stimulating bowel contractions, but unfortunately also affects the brain. I feel spacey, emotionally vulnerable, anxious and sometimes depressed. Prucalopride is not covered by my insurance, but I was able to obtain it affordably through an overseas pharmacy."

"I found it to be the most helpful out of all medications tried. My dose is 2 mg, it does not help with obstruction but with regular constipation, it is the first time in years I have had a fully formed soft stool. Make sure you eat before taking it. I did get very bad nausea taking it on an empty stomach."

"It took around a week to get over the side effects. I’m on 1mg of Prucalopride and I experienced minor headaches (treated with coffee) After about 2 weeks, I started having multiple bowel movements and things couldn’t be any better. I finally feel like I have a normal toilet life. I’ve read many reviews and feel like people who start off on 2mg experience far worse side effects but it’s extremely worth it once the side effects subside."

"Hi. I’ve had constipation since I was a child. Post-birthing two kids, at age 47, and as I enter perimenopause, it’s gotten notably worse. I have a diagnosed slow colon. The only thing that’s worked for me is Chinese herbs daily and acupuncture monthly. Cost prohibitive, though. Hopeful for this drug, but it steals my sleep, makes my teeth and head hurt, and makes me anxious and depressed when I take it for longer than a week. I have taken it two days in a row for relief, and I’m a mess. Anxious and sleepless. So, it’s not for everyone. Back to senna."

"I started on 1mg a day for a week. They gave me a slight headache but made me feel so, so sick for about 4 days. I couldn’t get out of bed for one of those days. So I took half a mg in the morning + in the evening to help ease into them. I also experienced strong waves in my stomach. But once I got through the initial shock to my system, they are working like a charm. I’m glad I personally persisted with them."

"developed sever IBS- C following emergency gynae surgery. Apparently that can bring on IBS. 4 years of 6-12 laxido a day. Frequent pseudo obstructions. Linzess stopped working within a week. Started prucalopride 1mg for 2 weeks with some success- didn't stop my laxido but adjusted it as needed. upped to 2mg. now on 4 laxido a day to keep it very soft. Have much fewer problems now. Also for 4 years couldn't eat and veg or fruit as I obstructed as my bowel couldn't move it, Now I have 2-4 portions a day. Am I cured, NO, but this is a big improvement and my quality of life has greatly improved. I have been on it about 6 months. I must say it appears to work more month by month, I am now starting to have a few days on 3 laxido. I suggest having some osmotic laxatives if your Constipation is very bad, along with prucalopride. It isn't contraindicated in most people, but check with your dr first. my dr told me to stop initially but I did it with linzess and paid for months after."

"I was given samples of Motegrity because I have pelvic floor dysfunction, IBS-C, gastritis and a bit of gastric emptying delay. I wanted this to work so badly! The first day produced a perfect BM, but I have migraines and thought one was coming on all day. The next day, BM again, but I started having anxiety over dying! What? Depression hit the third day. I’m done! Going to just take senna. Nothing else works."

"I've suffered from severe constipation for years nothing has ever really helped me. I took prucalopride once around a month and a half ago and had a really bad headache, nausea, sweats, and abdominal pain and couldn't get off the toilet all night. I took it again around 2 weeks later and had the same side effects but didn't work for me as well as it didn't the first time around. I started taking it again last night along with 2 other laxatives I was told to take and my god I haven't got off the toilet since! Side effects have not been as bad for me this time around the only problem is my stomach is rumbling so much but I have no appetite! I'm hoping they will calm down soon and my appetite will come back."
