What is pozelimab?

Pozelimab is for use in adults and children at least 1 year old to treat complement hyperactivation, angiopathic thrombosis, protein losing enteropathy (CHAPLE) syndrome.

Pozelimab may also be used for purposes not listed in this medication guide.

Pozelimab side effects

Get emergency medical help if you have signs of an allergic reaction: hives, difficult breathing, swelling of your face, lips, tongue, or throat.

Read the Patient Wallet Card about the symptoms of meningococcal or other infections and learn what symptoms to watch for. Keep the card with you at all times.

You should not use pozelimab if you have a meningococcal infection.

How is pozelimab given?

Follow all directions on your prescription label and read all medication guides or instruction sheets.

Pozelimab is injected into a vein and also under the skin by a healthcare provider.

You will be watched for a short time to make sure you do not have an allergic reaction.

You may need medical tests.

Pozelimab dosing information

Usual Adult Dose for CHAPLE Syndrome:

Initial dose: Day 1: 30 mg/kg by intravenous infusion over a minimum of 1 hour
Maintenance dose: Day 8 and thereafter: 10 mg/kg subcutaneously once a week
Maximum dose: 800 mg once a week

Comments:
-According to Advisory Committee on Immunization Practices (ACIP) guidelines, meningococcal vaccination should be completed at least 2 weeks before administering the first dose of this drug.
-The maintenance dosage may be increased to 12 mg/kg once a week if there is insufficient clinical response after at least three weekly doses.
-Doses greater than 400 mg require two injections.
-Do not exceed the maximum infusion rate of 1000 mg/hour.
-Monitor for 30 minutes after completing the infusion.
-This drug has not been studied in the geriatric population.

Use: For the treatment of adult patients with CD55-deficient protein-losing enteropathy (PLE), also known as CHAPLE disease

Usual Pediatric Dose for CHAPLE Syndrome:

1 year or older:
Initial dose: Day 1: 30 mg/kg by intravenous infusion over a minimum of 1 hour
