Usual Adult Dose for Hereditary Amyloidosis

45 mg subcutaneously once a month

Comments:

• Administer this drug into the abdomen or upper thigh region.
• Health care providers or caregivers can also administer injections in the back of the upper arm.

Use: For the treatment of the polyneuropathy of hereditary transthyretin-mediated amyloidosis

Renal Dose Adjustments

Mild to moderate renal dysfunction (estimated GFR 30 to less than 90 mL/min/1.73 m2): No adjustment recommended
Severe renal dysfunction or ESRD (estimated GFR less than 30 mL/min/1.73 m2): Data not available

Liver Dose Adjustments

Mild liver dysfunction (total bilirubin up to 1 times the upper limit of normal [1 x ULN] and AST greater than 1 x ULN, or total bilirubin greater than 1 to 1.5 x ULN and any AST): No adjustment recommended

Administration advice:

• Before starting therapy, patients and/or caregivers should be trained on proper preparation and administration.
• The autoinjector should be removed from the refrigerator 30 minutes before injection and allowed to warm to room temperature. Do not use other warming methods.
• Inspect drug for particulate matter and discoloration before administration.
• Do not use if cloudiness, particulate matter, or discoloration is observed.
• If a dose is missed, administer it as soon as possible and resume dosing from the date of the most recently administered dose.

Storage requirements:

• Store refrigerated at 2C to 8C (36F to 46F) in the original carton, protected from light.
• If needed, this drug can be stored at room temperature (up to 30C [86F]) in the original carton for up to 6 weeks after which, it should be discarded.
• Do not freeze or expose to heat.

Patient advice:

• Read the US FDA-approved patient labeling (Patient Information and Instructions for Use).
• The recommended daily allowance of vitamin A should be taken, as this drug may decrease serum vitamin A levels.
• Notify your health care provider if you experience ocular symptoms suggestive of vitamin A deficiency (e.g., night blindness, dry eyes).
• Notify your health care provider if you are pregnant or plan to become pregnant during treatment with this drug.
