GENERALLY AVOID: Coadministration with potent or moderate inducers of CYP450 3A4 may significantly decrease the plasma concentrations of daridorexant, which is primarily metabolized by the isoenzyme. Per physiologically based pharmacokinetic (PBPK) analysis, concomitant use of rifampin, a potent CYP450 3A4 inducer, decreased daridorexant systemic exposure (AUC) by more than 50%. When a 50 mg daridorexant dose was coadministered with multiple 600 mg doses of efavirenz, a moderate CYP450 3A4 inducer, daridorexant peak plasma concentration (Cmax) and AUC decreased by 40% and 60%, respectively. Reduced efficacy of daridorexant may occur.
MANAGEMENT: According to the manufacturer, concomitant use of daridorexant with potent or moderate CYP450 3A4 inducers should be avoided.

You should preferably avoid consumption of grapefruit and grapefruit juice while taking encorafenib. Grapefruit and grapefruit juice can significantly increase the blood levels of encorafenib. This may increase the risk of serious side effects such as bleeding complications, eye and vision problems, liver problems, irregular heart rhythm, and development of new skin cancers. Talk to your doctor if you have any questions or concerns. It is important to tell your doctor about all other medications you use, including vitamins and herbs. Do not stop using any medications without first talking to your doctor.
daridorexant food
Applies to: Quviviq (daridorexant)
Do not consume grapefruit, grapefruit juice, or any supplements that contain grapefruit extract during treatment with daridorexant unless directed otherwise by your doctor. Grapefruit juice can increase the blood levels of daridorexant. This may increase the risk of side effects such as somnolence, fatigue, abnormal sleep behaviors, worsening of depression or suicidal thoughts, or headache. Talk to your doctor if you have any questions or concerns. It is important to tell your doctor about all other medications you use, including vitamins and herbs. Do not stop using any medications without first talking to your doctor.
