GENERALLY AVOID: Coadministration of alpelisib with potent inducers of CYP450 3A4 may significantly reduce the plasma concentrations of alpelisib. The proposed mechanism, based on in vitro data, is increased clearance due to induction of CYP450 3A4, which is one of the routes of metabolism of alpelisib. The therapeutic efficacy of alpelisib may be reduced; however, clinical data are lacking.
MANAGEMENT: According to the manufacturer, concomitant use of alpelisib with potent CYP450 3A4 inducers should generally be avoided. Alternative therapeutic agents with less enzyme induction potential should be considered.

You should preferably avoid consumption of grapefruit and grapefruit juice while taking encorafenib. Grapefruit and grapefruit juice can significantly increase the blood levels of encorafenib. This may increase the risk of serious side effects such as bleeding complications, eye and vision problems, liver problems, irregular heart rhythm, and development of new skin cancers. Talk to your doctor if you have any questions or concerns. It is important to tell your doctor about all other medications you use, including vitamins and herbs. Do not stop using any medications without first talking to your doctor.
alpelisib food
Applies to: Vijoice (alpelisib)
Food significantly enhances the absorption of alpelisib. You should take each dose of alpelisib at the same time each day with a meal. Taking it on an empty stomach may lead to inadequate blood levels and reduced effectiveness of the medication. Talk to your doctor if you have questions or have difficulty taking alpelisib with food. It is important to tell your doctor about all other medications you use, including vitamins and herbs. Do not stop using any medications without first talking to your doctor.
