GENERALLY AVOID: Coadministration with potent inducers of CYP450 3A4 may significantly decrease the plasma concentrations of istradefylline, which is primarily metabolized by CYP450 3A4 and 1A1. In study subjects, administration of istradefylline with the potent CYP450 3A4 inducer rifampin (600 mg daily for 20 days) led to a reduction in the peak plasma concentration (Cmax) and total systemic exposure (AUC(inf)) of istradefylline by 45% and 81%, respectively, when compared with istradefylline administered alone.
MANAGEMENT: The use of istradefylline in combination with potent CYP450 3A4 inducers is not recommended. Alternative agents with no or minimal CYP450 3A4 induction potential are recommended whenever possible.

You should preferably avoid consumption of grapefruit and grapefruit juice while taking encorafenib. Grapefruit and grapefruit juice can significantly increase the blood levels of encorafenib. This may increase the risk of serious side effects such as bleeding complications, eye and vision problems, liver problems, irregular heart rhythm, and development of new skin cancers. Talk to your doctor if you have any questions or concerns. It is important to tell your doctor about all other medications you use, including vitamins and herbs. Do not stop using any medications without first talking to your doctor.
istradefylline food
Applies to: Nourianz (istradefylline)
Smoking tobacco may reduce the blood levels and effects of istradefylline. Contact your doctor if istradefylline is not adequately controlling your symptoms. It is best to avoid smoking during istradefylline therapy. Talk to a healthcare professional if you have any questions or concerns. If you start smoking or undergo smoking cessation, your doctor may need to monitor you more closely to determine if you need a dose adjustment of istradefylline. Your doctor may already be aware of the risks and may have taken appropriate precautions including adjusting your dose and monitoring you closely for any potential complications. It is important to tell your doctor about all other medications you use, including vitamins and herbs. Do not stop using any medications without first talking to your doctor.
