MONITOR: Coadministration with inhibitors of uridine diphosphate glucuronosyltransferase (UGT) 2B7 may increase the plasma concentrations and effects of capivasertib, which is a substrate of the isoenzyme. Based on physiologically based pharmacokinetic models, concomitant use with UGT2B7 inhibitor, probenecid is predicted to cause an increase in capivasertib systemic exposure (AUC) of 37% over a dosing cycle. However, the clinical significance of this effect is unknown.

MANAGEMENT: Caution is advised if capivasertib is used concurrently with medications that are inhibitors of UGT2B7. Clinical and laboratory monitoring may be appropriate for capivasertib whenever these drugs are added to or withdrawn from therapy. The prescribing information for the concomitant medication(s) may be consulted for further guidance.

Consumption of grapefruit, grapefruit juice, and supplements that contain grapefruit should be avoided during treatment with capivasertib as they may increase the blood levels of capivasertib. This may increase the risk of side effects such as high blood sugar levels, diarrhea, skin reactions, nausea, vomiting, tiredness, and changes in certain blood tests. Talk to your doctor if you have any questions or concerns. It is important to tell your doctor about all other medications you use, including vitamins and herbs. Do not stop using any medications without first talking to your doctor.

Alcohol can increase the nervous system side effects of valproic acid such as dizziness, drowsiness, and difficulty concentrating. Some people may also experience impairment in thinking and judgment. You should avoid or limit the use of alcohol while being treated with valproic acid. Do not use more than the recommended dose of valproic acid, and avoid activities requiring mental alertness such as driving or operating hazardous machinery until you know how the medication affects you.
