"I was put on prucalopride to treat moderate gastroparesis. I initially started on 1 mg and within a few weeks I became severely depressed, crying all the time and had suicidal ideation. I dropped the dose to 0.5 mg after reading the information sheet. My gastroparesis got worse, so I was told to go back up to 1 mg, and again within about 2-3 weeks I had severe depression and suicidal thoughts. I reduced the dose back to 0.5 mg, and within a few days the fog had lifted. I have the same reaction to tramadol, which also works in a similar way on serotonin receptors. Never again. Stay away from this drug if you have a history of mental health conditions, and if you're going to try, get someone to watch you closely."

For Chronic Idiopathic Constipation "Started taking 2 mg Resolor 10 days ago before bed, and apart from a bad headache the first night/morning, no other side effects to date. Some improvement in natural bowel movement, but the consultant recommended 2 weeks before seeing results. Fingers crossed."

"I have had irritable bowel syndrome with constipation (IBS-C) for around 10 years, which has worsened since having kids. I took the 2 mg dose as prescribed by my gastro consultant before breakfast this morning, and within 45 minutes, I was in a lot of pain and had diarrhea, headaches, and dizziness. The effects eventually wore off after around 4 hours, but they were pretty extreme. After having dinner tonight, I had extreme pain in my lower gut that felt like stabbing soreness. I also had gas and bowel movements and couldn't stand up straight from the pain. Not nice at all and will not be taking it anymore."

For Chronic Idiopathic Constipation "I have tried a variety of medications. Lubiprostone (Amitiza) works the best, but it still wasn't enough. I also still use Miralax almost daily and a stool softener once or twice a week. Adding prucalopride to these has helped to give me regular, predictable bowel movements. I take 1/2 pill (1 mg) once a day or every other day. So I'm pleased with how it helps with constipation. But I am minimizing the dose because it affects me like an SSRI antidepressant, which I've been on before and really dislike. Like SSRIs, prucalopride increases serotonin, which is helpful in stimulating bowel contractions, but unfortunately also affects the brain. I feel spacey, emotionally vulnerable, anxious, and sometimes depressed. Prucalopride is not covered by my insurance, but I was able to obtain it affordably through an overseas pharmacy."

For Chronic Idiopathic Constipation "I’ve had constipation since I was a child. Post-birthing two kids, at age 47, and as I enter perimenopause, it’s gotten notably worse. I have a diagnosed slow colon. The only thing that’s worked for me is Chinese herbs daily and acupuncture monthly. Cost prohibitive, though. Hopeful for this drug, but it steals my sleep, makes my teeth and head hurt, and makes me anxious and depressed when I take it for longer than a week. I have taken it two days in a row for relief, and I’m a mess. Anxious and sleepless. So, it’s not for everyone. Back to senna."

For Chronic Idiopathic Constipation "It took around a week to get over the side effects. I’m on 1 mg of prucalopride and I experienced minor headaches (treated with coffee). After about 2 weeks, I started having multiple bowel movements, and things couldn’t be any better. I finally feel like I have a normal toilet life. I’ve read many reviews and feel like people who start off on 2 mg experience far worse side effects, but it’s extremely worth it once the side effects subside."

For Chronic Idiopathic Constipation "I suffer from a range of gastro issues, alongside a damaged ileum intestinal valve due to an autoimmune disease. I have one of the most 'sluggish' bowels my GI doctor has ever seen. Managing my condition has always been difficult, diet can only do so much. Laxatives make me sick and ruin my gut. Then came prucalopride. It works without fail every time, and boy, does it work. It doesn't make me feel sick like other laxatives and doesn't cause that feeling of 'chemical gut.' It does, of course, cause some fluid swelling while it works. But once the 'deed is done,' I feel like a new person. I don't recommend taking it at night, however, personally, that causes some really uncomfortable stomach aches. You want to take it on an empty stomach or before food to have the best effect and to get it 'over and done with' so you can get on with your day (be near a toilet!)."

For Chronic Idiopathic Constipation "I was on Resolor 1 mg for 3 months. It has been helpful for regular bowel movements. However, on 5th June, I developed for the first time a supraventricular tachycardia with a heartbeat of an average of 130. Now I have stopped taking it. I need views on whether my SVT was caused by Resolor 1 mg."

For Chronic Idiopathic Constipation "I started on 1 mg a day for a week. They gave me a slight headache but made me feel so, so sick for about 4 days. I couldn’t get out of bed for one of those days. So I took half a mg in the morning + in the evening to help ease into them. I also experienced strong waves in my stomach. But once I got through the initial shock to my system, they are working like a charm. I’m glad I personally persisted with them."

For Chronic Idiopathic Constipation "Developed severe IBS-C following emergency gynae surgery. Apparently, that can bring on IBS. 4 years of 6-12 Laxido a day. Frequent pseudo-obstructions. Linzess stopped working within a week. Started prucalopride 1 mg for 2 weeks with some success - didn't stop my Laxido but adjusted it as needed. Upped to 2 mg. Now on 4 Laxido a day to keep it very soft. Have many fewer problems now. Also, for 4 years, couldn't eat any veg or fruit as I obstructed, as my bowel couldn't move it. Now I have 2-4 portions a day. Am I cured? No, but this is a big improvement, and my quality of life has greatly improved. I have been on it about 6 months. I must say it appears to work more month by month, I am now starting to have a few days on 3 Laxido. I suggest having some osmotic laxatives if your constipation is very bad, along with prucalopride. It isn't contraindicated in most people, but check with your doctor first. My doctor told me to stop initially, but I did it with Linzess and paid for months after."

For Chronic Idiopathic Constipation "I was given samples of Motegrity because I have pelvic floor dysfunction, IBS-C, gastritis, and a bit of gastric emptying delay. I wanted this to work so badly! The first day produced a perfect BM, but I have migraines and thought one was coming on all day. The next day, BM again, but I started having anxiety over dying! What? Depression hit the third day. I’m done! Going to just take senna. Nothing else works."

For Chronic Idiopathic Constipation "At first, I was excited because I have bloated for a long time, and I started to fart after an hour. Then suddenly it starts working and I have to run to the toilet and have this horrible diarrhea and palpitations to the extreme. My stomach growls nonstop and I even stay in the toilet for an hour... For Chronic Idiopathic Constipation "I've suffered from severe constipation for years, nothing has ever really helped me. I took prucalopride once around a month and a half ago and had a really bad headache, nausea, sweats, and abdominal pain and couldn't get off the toilet all night. I took it again around 2 weeks later and had the same side effects but didn't work for me as well as it did the first time around. I started taking it again last night along with 2 other laxatives I was told to take and my god, I haven't gotten off the toilet since! Side effects have not been as bad for me this time around, the only problem is my stomach is rumbling so much, but I have no appetite! I'm hoping they will calm down soon, and my appetite will come back."

It worked amazingly in the beginning, but now it doesn't really work because I have to drink 2 laxatives every day. I have no side effects. For Chronic Idiopathic Constipation "I was given Resolor/prucalopride 2 mg to try to help my slow bowel system from gastroparesis. I had read the reviews on here, so I decided to only take half the pill, 1 mg. I took it with water around 11 pm, so this way if it did well, I could sleep. I laid down and was on my phone just relaxing when about 40 minutes later I started feeling my stomach gurgle. I got up and went to the restroom. Sure enough, it already gave me a bowel movement. It didn't end there. I was starting to feel nauseous. I paced around because my stomach was becoming so upset there was no way I could lay down and just fall asleep. I ended up throwing up all of my food from that day plus diarrhea at the same time! It was awful! I'm jealous of all the people who've had the great experience with this pill, but I'll NEVER take it again. I was up until 5 am sick. I've laid in bed all day because I still feel sick. If you try this, take as little as possible the first time! You'll be grateful you did if it goes bad!"

Prucalopride for Chronic Idiopathic Constipation "I wanted to share my story as I just took my first dose of this medication tonight. I'm crossing my fingers because nothing else has been effective, and I am currently down to 107 pounds at a height of 5'5". During the lockdown, I was exposed to toxins in a home I had recently moved into. After four months of being there, I experienced a wide range of symptoms, including severe gastric emptying delay and dysfunction in my colon. Whenever I eat, I suffer from severe consequences. At the moment, I'm on my third round of Xifaxan, which costs $2500 for each two-week treatment, and my gastroenterologist suspects it might be small intestinal bacterial overgrowth (SIBO) as well. Xifaxan helps with reducing inflammation and bloating, but unfortunately, my colon isn't improving. Currently, the only thing I am able to eat without major issues is eggs, and even then, I have to use a daily enema. I also have eosinophilic esophagitis (EOE) and have undergone several surgeries involving scopes, biopsies of the esophageal area, and laparoscopic exploratory surgery. As I enter my third year with these conditions, they are getting worse."

"I am on the fourth day of 1 mg for gastroparesis, and it is bad! Now I am normally sick every day with multiple illnesses, but this is horrible. On the 1st day, I took it, and within 1 hour, massive headache, severe watery diarrhea like 10 times in one hour, heart pounding so fast, had to take Gravol. I have been unable to even leave my house for 4 days now from these side effects. My doctor thought it would improve my GERD, absolutely did not! I'll try one more day and then stop. I would only use this if it's a life-or-death situation! I normally will have a regular bowel movement from a cup of strong coffee without these awful side effects!"
