12Next ..Centocor, Inc., and Schering-Plough recently announced the approval of Remicade, their chimeric monoclonal antibody to TNF-alpha, for the treatment of moderate to severe plaque psoriasis by the European Union. Remicade was approved for use in adult patients who otherwise failed or were intolerant of conventional systemic therapies including cyclosporine, methotrexate, and PUVA.

The European Union based its approval of Remicade for psoriasis on data obtained from 2 clinical trials: Study of Psoriasis with Infliximab Induction Therapy (SPIRIT) and European Infliximab for Psoriasis Efficacy and Safety Study (EXPRESS). These studies were multicenter randomized double-blinded, placebo-controlled trials that evaluated the efficacy of Remicade in patients with 10% body surface area affected and a PASI score of 12. The primary endpoint in the trials was PASI 75 from baseline to week 10. Both studies demonstrated that greater than 80% of patients receiving Remicade 5 mg/kg at weeks 0, 2, and 6 achieved a PASI 75. More than half the patients in each study achieved a PASI 90. Currently, Remicade is approved for psoriatic arthritis, rheumatoid arthritis, ankylosing spondylitis, and Crohn's disease in the US.
