If you started taking Tri-Sprintec on day 1 (the first day of your period) you will have immediate contraceptive protection and on any other day you will need to wait until you have taken 7 consecutive active tablets.
So in answering your question, regardless of when you started, as long as you have followed your doctors instructions and taken the tablets as directed you will be protected from pregnancy.
