By what mechanism does the rosiglitazone cause an increased risk of MI but pioglitazone does not have this effect?
Studies have suggested that Rosiglitazone may increase the risk of heart attack by as much as 43% . In some countries it has been taken off the market and in others it is only available under significant restrictions. Patients should be advised of alternative options.
Pioglitazone has a cardiovascular safety profile that compares favorably with rosiglitazone. However, pioglitazone has subsequently been found to be associated with bladder tumors and has been withdrawn in some countries.
