450mg/day is a rather large dose. However, original Effexor was tested and approved up to 375mg/day. Effexor XR maximum labeled dose is 225mg/day. But, that's because there were only studies submitted up to 225 on the XR. Theoretically, since it's XR you could take once daily. However, with the larger dose and the fact that venlafaxine has a short half-life, it is probably better to divide it up to lower peak concentrations. Most antidepressants don't show much added benefit beyond the standard dose. Effexor is different in that it does seem to have an increasing dose-response up to fairly high levels. It is sometimes used in these high doses when other options fail. The main side effects you may notice get worse are increased blood pressure/heart rate (just monitor it), increased sweating, maybe a little nausea or insomnia until you adjust.
