Description Of The Vaccine

Coliprotec® is an oral live vaccine, containing naturally avirulent Escherichia coli bacteria, recommended for the vaccination of healthy weaned pigs of 17 days or more as an aid in the prevention of post-weaning diarrhea (PWD) caused by F4(K88)-positive Escherichia coli. Clinical studies confirmed that Coliprotec® vaccine significantly reduced intestinal colonization of virulent enterotoxigenic Escherichia coli (ETEC), diarrhea, and accumulation of fluid in the intestines after a challenge.

Precautions

- Store refrigerated at 2-8°C (35-45°F). Do not freeze

- For oral use only

- Materials used in administering the vaccine must be free of antibiotic or disinfectant residue to prevent inactivation of Coliprotec®

- Do not use with antibiotics or other live vaccines for a minimum of 3 days prior to and after vaccination

- Remove antibiotics and disinfectants from drinking water for a minimum of 3 days prior to and after vaccination

- Use entire content when first opened

- Reconstituted vaccine should be used within 4 hours

- Do not vaccinate within 21 days before slaughter

Warning

Anaphylactic reactions may occur. Use epinephrine as antidote. Dispose unused product and empty containers according to local waste disposal requirements.

Coliprotec Dosage And Administration

The Coliprotec® vaccine is administered orally using one dose per pig. Three methods of administration are recommended, being 1) Individual oral administration using a syringe, 2) Conventional drinking water systems and 3) Automatic medicator (proportioner) watering system.

Directions For Administration

Individual administration using a syringe

Do not open or rehydrate the lyophilized vaccine until use. Water used for rehydration or for administration of the vaccine must be free of antibiotic or disinfectant residue to prevent inactivation. Remove antibiotics and disinfectants from drinking water for a minimum of 3 days prior to and after vaccination. Rehydrate the lyophilized bacterial culture in the vial using approximately 10 mL (200-dose format or less) or 20 mL (format of more than 200 doses) of water. Shake well. Transfer the vaccine in a sterile graduated sterile container. Rinse the bottle with another 10 mL of water and transfer in the same container. DILUTE THE VACCINE IN THE CONTAINER USING WATER TO OBTAIN A FINAL VOLUME OF 3 ML PER DOSE. Using a syringe, administer a 3-mL oral single dose of the vaccine preparation per pig.

Conventional drinking water system

Do not open or rehydrate the lyophilized vaccine until use. Water used for rehydration or for administration of the vaccine must be free of antibiotic or disinfectant residue to prevent inactivation. Remove antibiotics and disinfectants from drinking water for a minimum of 3 days prior to and after vaccination. Flush the watering system with untreated water. Withhold water for 1 to 2 hours immediately before vaccination. Ensure that pigs can drink in 4 hours by providing water space so that all the pigs can drink easily.

Determine the volume (mL) of water consumed by the number of treated pigs within 4 hours by measuring the volume of water pigs drink during 4 hours at the day prior the vaccination. If not available, please refer to the reference table below. Rehydrate the lyophilized bacterial culture in the vial using approximately 10 mL (200-dose format or less) or 20 mL (format of more than 200 doses) of water. Shake well. Transfer the vaccine in a sterile container. Rinse the vial with another 10 mL of tap water and transfer in the same container. Dilute the vaccine with approximately 75% of the volume of water consumed by the number of treated pigs within 4 hours. Add the bacterial suspension to the drinking water tanks for the appropriate number of pigs. The rehydrated culture should be consumed within 4 hours.

Automatic medicator watering system

Do not open or rehydrate the lyophilized vaccine until use. Water used for rehydration or for administration of the vaccine must be free of antibiotic or disinfectant residue to prevent inactivation. Remove antibiotics and disinfectants from drinking water for a minimum of 3 days prior to and after vaccination. Flush the watering system with untreated water. Withhold water for 1 to 2 hours immediately before vaccination. Ensure that pigs can drink in 4 hours by providing water space so that all the pigs can drink easily.

Determine the volume of stock solution to be prepared for the number of pigs to treat: Determine the volume (mL) of water used in the proportioner container (priory filled with a known volume of water) by the number of treated pigs within 4 hours at the day prior the vaccination. If not available, please refer to the reference table below. Select the volume of the proportioner for 30 mL of stock solution per 4 litres of water (1 oz per gallon). Many proportioners are factory set at 1:128 to deliver 1 oz per gallon of water.

Prepare the stock solution of culture: Use the number of doses for the number of pigs to treat. Rehydrate the lyophilized bacterial culture in the vial using approximately 10 mL (200-dose format or less) or 20 mL (format of more than 200 doses) of water. Shake well. Transfer the vaccine in the proportioner container. Rinse the vial with another 10 mL of water and transfer in the same container. Add the appropriate volume of water to reach the determined volume of stock solution to be prepared for the number of pigs to treat in 4 hours. Mix well. Insert the tube of the proportioner into the stock solution (vaccine preparation) and open the water system. The rehydrated culture should be consumed within 4 hours.
