Interactions between ranitidine and Erlotinib (erlotinib)

Moderate Drug-Drug Interaction ranitidine and erlotinib (Moderate Drug-Drug)

GENERALLY AVOID: Concurrent administration of agents that increase gastric pH, such as H2 antagonists and proton pump inhibitors, may decrease the oral bioavailability of erlotinib and reduce its concentrations in plasma. The solubility of erlotinib decreases when the pH is over 5, resulting in reduced absorption. After coadministration of omeprazole, the area under the concentration-time curve (AUC) of erlotinib decreased by 46% and the maximum plasma concentration (Cmax) decreased by 61%. Time to maximum concentration (Tmax) and half-life did not change. The clinical significance is unknown.

MANAGEMENT: The concomitant use of erlotinib with proton pump inhibitors or H2 antagonists should be avoided.
