MONITOR: Coadministration with metformin may decrease the plasma concentrations of trospium. The mechanism of interaction has not been established. In 44 healthy subjects, administration of trospium chloride extended-release 60 mg once daily in combination with metformin immediate-release 500 mg twice daily reduced the steady-state mean trospium peak plasma concentration (Cmax) by 34% and systemic exposure (AUC) by 29%. The clinical relevance of these changes is unknown. Whether metformin at higher dosages may have a greater effect on trospium pharmacokinetics has not been investigated. Trospium chloride did not significantly affect the steady-state pharmacokinetics of metformin.
MANAGEMENT: Patients should be monitored for potentially diminished efficacy of trospium chloride during coadministration with metformin.
