ritonavir ⇔ enfuvirtide
Applies to: Norvir (ritonavir) and Fuzeon (enfuvirtide)
Coadministration with ritonavir and/or saquinavir may slightly increase the plasma concentrations of enfuvirtide. The mechanism of interaction is unknown. According to the manufacturer, ritonavir (200 mg every 12 hours for 4 days) increased the steady-state peak plasma concentration (Cmax), area under the concentration-time curve (AUC) and trough concentration (Cmin) of enfuvirtide (90 mg twice a day) by 24%, 22% and 14%, respectively, in 12 HIV-positive subjects. Saquinavir-ritonavir (1000 mg-100 mg every 12 hours for 4 days) increased enfuvirtide AUC by 14% and Cmin by 26% but did not affect Cmax. These alterations are not considered to be clinically significant.
