saquinavir ⇔ etravirine
Applies to: Fortovase (saquinavir) and Intelence (etravirine)
Coadministration with saquinavir/ritonavir may decrease the plasma concentrations of etravirine. The mechanism of interaction has not been described but may involve ritonavir induction of CYP450 2C19 and 2C9, of which etravirine is a substrate. In 14 study subjects administered etravirine with saquinavir/ritonavir (1000 mg/100 mg) twice a day, etravirine peak plasma concentration (Cmax), systemic exposure (AUC) and trough plasma concentration (Cmin) decreased by 37%, 33% and 29%, respectively. These changes are similar to those observed when etravirine is coadministered with darunavir/ritonavir. Because all subjects in Phase 3 trials of etravirine received darunavir/ritonavir as part of the background regimen and etravirine exposures from these trials were determined to be safe and effective, etravirine and saquinavir/ritonavir may be coadministered without any dosage adjustments.
