I have been taking Amaryl (glmapride) for 12 years. Does your body get use to it and should I change or can I take it forever?

I have known people to take it for a long time. If and when your blood glucose levels aren't controlled, or if you experience side effects from Amaryl then your doctor will probably look at upping your dose or putting you on some other medicine.
