I realize this answer is very late but ... I am sure Emla will not affect a urine sample.
The elimination half life of both lidocaine and prilocaine is about 150minutes (max). It takes 5.5 x elimination half lifes for a medicine to be out of ones system so Emla should be out of your system in 825 mins i.e. approximately 14 hours (5.5 x 150 mins). So if you had a urine test after about 15 hours lidocaine and prilocaine would have been cleared from your system.
